1) Sign up with a real email address - avoid using free services like Gmail or Hotmail when possible because sometimes the moderators automatically mark these as spam since they are heavily used by automation software.  You want to appear human (because you are, right???)

2) After you sign up and confirm through your email wait 5-10 days before placing your link in your profile page.  Many sites will delete brand new members that just sign up and post a link on day 1.  By waiting a few days you are soon forgotten about and you won't show up on the 'newest members' page anymore which is where the moderators and admins usually check through profiles.

3) Once you have the URL for your backlink you need to make sure that Google actually finds the page and indexes it.  If Google never finds the page then you won't get credit for the backlink.  From my experience Google finds about 9% of profile links on their own.  That means if you build 100 profile links then the chances are they will only find about 9 of them.

You can easily increase this to about 80-90% with a few simple steps.  You could use a service like Backlinks Indexer, build social bookmarks with Senuke X, or do some manual link building from Digg, Reddit, Stumbleupon to help get them indexed.
